摘要:
目的:观察慢性快速眼动相睡眠剥夺(chronic REM sleep deprivation,CSD)对大鼠学习记忆能力以及海马、前额皮质、下丘脑5-HT1A受体蛋白表达变化的影响。 方法:成年雄性SD大鼠经过筛选后随机分为CSD组(n=15)、大平台实验对照TC组(n=12)和空白对照BC组(n=4)。采用改良多平台水环境方法建立大鼠慢性REM睡眠剥夺模型,利用Morris水迷宫、自主活动箱检测CSD后大鼠学习记忆功能变化,Western Blot法分析CSD对大鼠海马、前额皮质、下丘脑5-HT1A受体蛋白表达水平的影响。 结果:与BC组和TC组相比,CSD组大鼠毛色无光泽、疲惫且易激惹;自睡眠剥夺3d起CSD组大鼠体重明显减轻(P值均<0.01)。CSD组Morris水迷宫潜伏期在睡眠剥夺第7、14、21d时间点与TC组相比明显延长(P值均<0.01),在第21d时间点与BC组相比也明显延长(P值<0.01);睡眠剥夺21d后CSD组自主活动箱中央区活动速度与TC组相比明显减慢(P值<0.05),与BC组相比差异没有统计学意义(P值>0.05)。睡眠剥夺21d后CSD组不同脑区内5-HT1A受体蛋白表达较TC、BC组增加(P值均<0.05);而TC组与BC组相比,学习记忆能力、各脑区5-HT1A受体蛋白表达差异均无统计学意义(P值>0.05)。 结论:慢性REM睡眠剥夺导致大鼠学习记忆能力下降,可能与脑区中5-HT1A受体调节大鼠睡眠有关。
Objective: To investigate the effect of chronic REM sleep deprivation on rats learning and memory, and the change of 5-HT1A receptor in rats hippocampus(HIP), prefrontal cortex (PFC) and hypothalamus(HTH). Methods: Male SD rats were randomly divided into chronic REM sleep deprivation (CSD, n=15), big platform treatment control (TC, n=12) and blank control (BC, n=4) groups after screening experiment. The sleep deprivation model was established by modified multiple platform method. Learning and memory function were detected by Morris water maze and open field test pre and post sleep deprivation. The effect of chronic REM sleep deprivation on 5-HT1A receptor protein expression in the HIP, PFC and HTH were quantitatively analyzed by Western Blot technique. Results: Compared with BC and TC groups, CSD group were exhausted and irritated, weight of CSD rats decreased significantly compared with TC and BC rats from 3d sleep deprivation to the end (P<0.01); For the Morris water maze, CSD rats latency was longer than TC rats 7, 14, 21d and BC rats 21d after sleep deprivation (P<0.01). CSD rats motion speed was more slowly in the open field test than TC rats (P<0.05), and no significant differences to BC rats (P>0.05). The protein expression of 5-HT1A receptor in 3 encephalic regions increased in CSD rats than the other two group rats (P<0.05). There were no significant differences in learning and memory function and protein expression of 5-HT1A receptor in 3 encephalic regions between BC and TC group. Conclusion: Chronic sleep deprivation exerts impaired effects on learning and memory, 5-HT1A receptor could be involved in modulating the dysfunction.
