摘要:
【摘要】 目的 研究幼年期应用右旋美托咪定对成年后的大鼠有认知功能的影响。方法(1)将60只幼年SD大鼠,随机分为两组,实验组术后予以右旋美托咪定,对照组术后不予以右旋美托咪定。大鼠成年后进行认知行为学研究,包括旷场实验,水迷宫,条件性恐惧记忆实验。提取各组大鼠海马组织,进行western blotting免疫组化,检测海马认知相关的分子通路变化。 结果 幼年期大鼠应用右旋美托咪定对大鼠成年后的空间记忆能力,场景记忆能力,声音恐惧记忆等认知能力没有明显的影响,对海马脑区认知相关分子通路也没有显著影响。结论 右旋美托咪定对大鼠大脑发育及远期认知功能无明显影响。
【Abstract 】Objective To study the influence of dexmedetomidine administration in infancy on cognitive function later in adulthood. Methods 60 infant SD rats were randomly divided into two groups, the control group did not apply dexmedetomidine, while the experimental group were used dexmedetomidine during and after surgery. We conduct cognitive behavior tests, including open field test, Morris water maze, fear conditioning memory experiment when the rats grow up. The expression of related signal proteins in hippocampus such as CaMKII, CREB and BDNF after dexmedetomidine administration was measured by western blotting and immunohistochemistry. Results Dexmedetomidine did not affect spatial memory, contextual memory and tone-dependent fear memory and other cognitive abilities in rats, and no significant difference was found on cognitive signal proteins in hippocampus between two groups. Conclusion Administration of dexmedetomidine does not have influence on the brain development and long-term cognitive function of rats.
