摘要:
【摘 要】 目的 探讨过氧化物酶体增殖物激活受体-γ(PPAR-γ) 在自发性高血压大鼠(SHR) 胸主动脉的表达及瑞舒伐他汀(ROS)的干预作用。方法 6周龄雄性SHR (n=16),随机分为SHR组(n=8)、ROS组(n=8);另选同龄雄性Wistar-Kyoto大鼠(WKY)作为对照组(n=8),喂养8周后抽取血液,处死,获取胸主动脉段,HE染色并计算机图像分析系统测量中膜厚度/管腔内径(M T/L D)、中膜面积/管腔面积(M A/L A),以双抗体夹心ABC-ELISA法检测血清IL-6、TNF-α、hs-CRP浓度,取主动脉组织以 Real-Time -PCR 及Western Blot检测PPAR-γ基因和蛋白表达水平。结果 SHR组血清IL-6,TNF-α,hs-CRP水平均明显高于WKY对照组;ROS组血清IL-6,TNF-α,hs-CRP浓度明显低于SHR组 (102.62±15.28 pg/ml vs 198.72±28.18pg/ml;8.92±2.48 pg/ml vs 16.32±3.32pg/ml;0.89±0.16 ng/ml vs 1.64±0.24ng/m1;P均<0.05);SHR组M T/L D 、M A/L A比WKY组显著增加(P<0.01),ROS组M T/L D 、M A/L A比SHR组明显下降(P<0.01);SHR组胸主动脉PPAR-γ基因和蛋白表达含量均明显高于WKY组(P均<0.01),ROS组胸主动脉PPAR-γ基因和蛋白表达含量均明显高于SHR组(P均<0.01)。结论 PPAR-γ可能与高血压血管重构有关,瑞舒伐他汀科可抑制炎症,促进血管中的PPAR-γ表达,改善血管重塑。

Methods SHR(n=16) were randomly divided into SHR (n=8) group and rosuvastatin(ROS) group(n=8) respectively,age-matched Wistar-Kyoto(WKY) (n=8) rats were selected as control group. After eight-weeks’ treatment,serum level of IL-6,TNF-α,hs-CRP were detected with ABC-ELISA.Media thickness /lumen diameter (MT/LD) and Media area/lumen area (MA/LA) of thoracic aorta were measured. The thoracic aortic PPAR-γ mRNA and protein expression were detected with Real-time-PCR and Western blot. Results ROS group was compared with SHR group, Serum level of IL-6,TNF-α,hs-CRP(102.62±15.28 pg/ml vs 198.72±28.18 pg/ml,8.92±2.48 pg/ml vs 16.32±3.32 pg/ml,0.89±0.16 ng/ml vs 1.64±0.24 ng/m1) in ROS group was significantly decreased(P<0.05).Compared with the SHR group,MT/LD and MA/LA of thoracic aorta in ROS group were significantly reduced (P<0.05);The expression of thoracic aorta PPAR-γ mRNA and protein in the ROS group was significantly higher than that of the SHR group(P<0.01).Conclusion PPAR-γ is likely to be related to vascular remodeling, and rosuvastatin could increase the expression of PPAR-γ and improve vascular remodeling in SHR.
