摘要:
目的:观察不同浓度(0,1,5,10,50,100,120μmol/L)的白藜芦醇(Resveratrol, Res)对体外培养大鼠心肌微血管内皮细胞(MMVECs)血管生成的影响及探讨其可能机制。方法:植块法培养并鉴定大鼠的MMVECs,取第二代细胞进行实验;并在加入不同浓度的Res处理后观察:1)其对MMVECs增殖、迁移和体外管腔样结构形成能力的影响;2)采用Western blot测定MMVECs中腺苷酸活化蛋白激酶(AMPK)和内皮型一氧化氮合酶(eNOS)磷酸化水平改变,评价其在Res促血管形成过程中的作用价值。结果:与对照组相比,Res呈呈剂量依赖性促进了内皮细胞的增殖、迁移和体外管腔形成,这一促血管新生过程伴随有AMPK和eNOS磷酸化程度的增加,其效应在浓度为100μmol/L时最显著。结论:Res呈剂量依赖性促进了大鼠MMVECs的体外血管新生,这一作用很可能与AMPK/eNOS信号通路的激活相关。
Objective: To study the effects of different doses of resveratrol (Res) on angiogenesis in MMVECs and its underlying mechanisms. Methods: Myocardial microvascular endothelial cells(MMVECs) were isolated and identified from 6-8 weeks’ Wistar rat . Only second passage were used in our study. After 2h incubated with different doses of resveratrol (0,1,5,10,50,100,120μmol/L), the proliferation, migration and tube-like structure formation ability of MMVECs were tested; The expression and phosphorylation of AMPK and eNOS after treated with resveratrol were measured with Western blot. Results: Compared with control, Res promote angiogenesis, including proliferation, migration and tube-like structure formation on MMVECs in a dose manner, which was closely associated with increased phosphorylation levels of both AMPK and eNOS,and the maximum effect being served at 100μmol/L. Conclusion: Res can dose-dependently promote angiogenesis of MMVECs possibly via activation of AMPK/eNOS signaling.
