摘要:
目的:观察反复给予氯胺酮对大鼠海马神经元p-cofilin表达的影响及其可能参与氯胺酮导致认知功能障碍的机制。方法:40只2月龄SD雄性大鼠随机分为两组:氯胺酮组(K组)和生理盐水组(C组)。氯胺酮组每天腹腔注射50mg/kg氯胺酮,连续7天;生理盐水组每天腹腔注射1ml/kg生理盐水,连续7天。2天后用morris水迷宫进行定位航行实验和空间探索实验,检测其学习记忆能力,并从氯胺酮组筛选出认识功能障碍模型组(M组)。采用Western blot法分析认知功能障碍模型组和生理盐水组大鼠海马内p-cofilin与cofilin的表达变化。结果:(1)氯胺酮组比生理盐水组平均逃逸潜伏期明显延长(P<0.05),穿越平台次数显著减少(P<0.05)。(2)与生理盐水组相比,认知功能障碍模型组大鼠海马内p-cofilin表达明显降低(P<0.05); 而cofilin总量表达无显著性差异(P >0.05)。结论:大鼠反复给予氯胺酮后,可以导致大鼠认知功能障碍,大鼠海马内p-cofilin降低。
Objective To investigate the effect of repeated ketamine administration on p-cofilin in rat hippocampus and their possible mechanism be involved in ketamine resulted in the cognitive dysfunction. Methods Forty 2 -month-old male SD rats were randomly divided into two groups : ketamine group (K group) and saline control group (C group).Rats in K group had been intraperitoneally injected with 50 mg/kg ketamine, and those in C group had been administrated with 1ml/kg saline for one week.After two days,Morris water maze test was adopted to explore the latency in place navigation and spatial probe ability. Through the morris water maze , the cognitive deficit model group (M group) was screened from the ketamine group. The expression of p-cofilin and cofilin in hippocampus of the cognitive deficit group and saline control group rats were analysed by Western blot. Results (1)In Morris water maze test, the latency in place navigation of K group was significantly longer than that of C group (P<0.05), and the spatial probe ability significantly decreased (P<0.05).(2)In western blot test, it was revealed that the expression of p-cofilin in M group was significantly lower than that in C group(P<0.05) ,while there was no significant difference in the of expression of cofilin between these two groups (P>0.05).Conclusion Repeated administration of ketamine in rats may cause cognitive dysfunction and reduce p-cofilin in rat hippocampus.
