摘要:
目的 研究CD105在标记腹膜透析腹膜新生血管中的价值。 方法 40只SPF级雄性SD大鼠按数字随机法分为正常组(假手术组)、尿毒症组、正常腹透组和尿毒症腹透组(2.5%、4.25%腹透液)(n=8)。腹透组SD大鼠每日经腹透管腹腔灌注4.25%葡萄糖透析液(10 ml/100g体重)一次共4至8周,各组大鼠均留取脏层腹膜\壁层腹膜\肝脏皱褶,进行电镜检测,观察内皮细胞形态及其连接。各组大鼠处死前,取脏层腹膜标本予CD105免疫荧光染色并计算血管数。采用Western印迹法在蛋白水平检测腹膜血管内皮生长因子-2型受体(VEGFR-2)的表达量,观察CD105标记的腹膜新生血管数和VEGFR-2蛋白表达量的关系。结果 与假手术组大鼠相比,尿毒症组大鼠12周的血尿素氮、血肌酐和24小时尿蛋白定量均明显升高(P<0.01),透析过程对实验动物的生化指标没有明显影响;尿毒症腹透组较正常组大鼠腹膜新生血管数量增多(12±4)/HP VS (1±1)/HP(P<0.01);CD105标记的新生血管数与VEGFR-2的检测结果显著相关(r=0.52,P<0.05)。结论 CD105可以作为腹膜透析腹膜新生血管较好的检测指标,为腹膜血管新生的评估、疾病严重程度的判断、治疗效果的评价,提供新的、有效的检测手段。
Objective: To investigate the association between CD105 and peritoneal angiogenesis in a uremic peritoneal dialysis(PD)rat model. Methods : Uremic (5/6 subtotal nephrectomy)rats were established and divided into non-PD, chronic renal failure, uremic+PD(2.5% or 4.25%) for 4 to 8weeks, normal+PD( 4.25%) for 4weeks. Standard PD solution was applied in the study. Rats undergone sham operation without PD process were used as control group. Vessel density of the peritoneum was detected and quantified with anti-CD 105 immunofluorescence staining.Expressive levels of VEGFR-2 (vascular endothelial growth factor-receptor 2) were examined in the peritoneum by Western blotting.Results: After subtotal nephrectomy, the rats developed increased serum creatinine and blood urea nitrogen (BUN), and polyuria as well as increased urinary loss of total protein. The non-PD group was characterized by no increased vessel density in the peritoneum compared with PD control group (1±1)/HP VS(12±4)/HP].Progressive angiogenesis was found in uremic+4.25%PD groups for 4 or 8weeks.Furthermore,expressive levels of VEGFR-2 keep high level in the PD group compared with the control (P<0.01),and such expressions were significantly higher in the PD groups as compared to normal+PD group (P<0.01). VEGFR-2 levels were positively correlated with new vessel density markded by CD105.(r=0.5178,P<0.05);Conclusions: Increased expression level of CD105 in peritoneum is positively correlated with peritoneal angiogenesis. CD105 may be a new biomark to stain the angiogenesis in the peritoneum.
