摘要:
目的:探讨在兔动脉粥样硬化斑块模型中PPAR-γ的表达水平以及可能作用。方法:25只雄性兔随机分为5组:正常饮食组;高脂饮食组(6周、10周);高脂饮食+球囊损伤手术组(6周、10周)。行HE染色观察动脉粥样斑块的病理变化,免疫组化法和Western Blot,以及realtime-PCR方法检测各组PPAR-γ蛋白和 mRNA表达情况,ELISA法测血清中肿瘤坏死因子—α、P选择素以及基质金属蛋白酶—9(4周、6周、8周、10周)水平。结果:高脂和高脂+球囊损伤各组,PPAR-γ的表达水平随着时间增加而增加(P<0.01),且高脂+球囊损伤组表达增强(P<0.01)。血清肿瘤坏死因子—α、P选择素以及基质金属蛋白酶—9在高脂组和高脂+球囊损伤各组中的水平明显都高于正常组(P<0.01),且在第10周较第8周水平降低(P<0.05)。结论:PPAR-γ在动脉粥样硬化斑块病变严重处表达增强,可以通过调控炎症反应,促进动脉硬化发生的作用。
Objective:To investigate the expression and the possible effect of peroxisome proliferator-activated receptor-gamma in atherosclerotic. Methods:A total amount of 25 male rabbits were randomly divided into 5 groups: control, high-fat diet(6th week、10th week), high-fat diet with balloon-injury(6th week、10th week). Alterations in tissue structure were assessed in artery tissue samples by using hematoxylin and eosin. Immunohistochemistry (IHC) and western blot analysis were used to examine PPAR-γ protein expression. The expression of PPAR-γ mRNA was determined by real-time quantitative PCR. Tumor necrosis factor alpha, P—selectin—major and matrix metalloproteinase—9 inflammatory factors(4th week, 6th week, 8th week, 10th week)involved in atherosclerosis were monitored during the experiment. Results: High in fat and high in fat + balloon injury group, the expression of PPAR-gamma as time increased (P < 0.01),and enhanced expression in fat + balloon injury group were Tumor necrosis factor alpha, P—selectin—major and matrix metalloproteinase— 9 in high-fat group and high-fat + balloon injury group were obviously higher than that of normal group, and compared with 8th weeks, 10th weeks levels were reduced (P < 0.05). Conclusion: The results of this study demonstrate that PPAR-γ may play role in the occurrence and development of atherosclerosis, which related to regulation the inflammation.
