摘要:
目的 本实验观察阿魏酸钠片早期干预由脱氢野百合碱诱导肺动脉高压比格犬的疗效。方法 分组 正常对照组n=6(溶剂0.1ml/kg二甲基甲酰胺+安慰剂);阿魏酸钠片组n=6(2mg/kg脱氢野百合碱+20mg/kg阿魏酸钠片);脱氢野百合碱组n=6(2mg/kg脱氢野百合碱+安慰剂)。药物干预前、造模4周后和给药4周后测量血流动力学参数,比格犬处死后取肺组织做H.E染色光镜观察。 结果 造模后4周阿魏酸钠片组、脱氢野百合碱组右心室压、肺动脉收缩压、肺动脉平均压力和肺血管阻力明显升高,心输出量明显下降,与正常对照组比较均具有明显差异(P<0.01)。给药四周后,阿魏酸钠片组与正常对照组比较无明显差异,但两组与脱氢野百合碱组比较均具有明显统计学差异(P<0.01)。H.E染色示脱氢野百合碱组肺小动脉内膜显著增厚和新生内膜形成,肺小动脉增生伴充血。阿魏酸钠片组肺小动脉内膜增厚较脱氢野百合碱组轻。正常对照组无上述改变。结论 脱氢野百合碱诱导比格犬肺动脉高压模型的形成,阿魏酸钠片早期干预能降低肺动脉压力,降低血管内膜增生。
Objective To access whether sodium ferulate is capable of attenuating pulmonary hypertension in beagles induced by dehydro monocrotaline(DHMC). Methods 18 healthy beagles were used, they were divided into three groups. Control group n=6 (0.1ml/kg dimethylformamide + placebo). Sodium ferulate group n=6(2mg/kg DHMC + 20mg/kg sodium ferulate). DHMC group n=6 (2mg/kg DHMC + placebo). Before drug intervention, 4 weeks after modeling and after 4 weeks of drugs , hemodynamic parameters were measured. Then the Beagles were killed and H.E staining was made in lung tissues. Results 4 weeks after modeling, both in sodium ferulate group and DHMC group the right ventricular pressure, pulmonary artery systolic pressure, mean pulmonary artery pressure and pulmonary vascular resistance were significantly increased and cardiac output decreased. There was a significant difference between control group and the two groups(P <0.01). After four weeks of drugs, there was no significant difference in sodium ferulate group and control group, but there was a significant difference in the two groups and DHMC group(P <0.01). H.E. staining showed in DHMC group pulmonary artery intima was significantly thickened and neointimal formation appeard. In sodium ferulate group pulmonary artery intima was not significantly thickened. No changes happened in control group. Conclusion DHMC could induce pulmonary hypertension in beagles and sodium ferulate could reduce pulmonary artery pressure and intimal hyperplasia in early stage.
