摘要:
目的 检测Notch1~4蛋白在肺腺癌组织中的表达,并分析其与临床特征的关系,探讨其临床意义。方法 搜集肺腺癌病例,调查其分化程度、肿瘤大小、淋巴结转移和临床分期等参数;采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结法(免疫组织化学SP法)分别检测腺癌组织及其癌旁组织中Notch1~4蛋白的表达,应用Image-Pro Plus 6.0图像分析软件进行图像检测,并分析其与临床病理因素的关系。结果 Notch1、2、3蛋白在肺腺癌在男性患者中的IOD值显著高于女性(P值均<0.05);在TNM分期Ⅱ、Ⅲ期患者中的IOD值均显著高于TNM分期Ⅰ期患者(P值均<0.05),在TNM分期Ⅲ期患者中的IOD值显著高于TNM分期Ⅱ期患者(P值均<0.05);在有淋巴结转移患者中的IOD值显著高于无淋巴结转移的患者(P值均<0.05);在有远处转移患者中的IOD值显著高于无远处转移的患者(P值均<0.05);在不同性别、年龄、分化程度、肿瘤大小的患者中的IOD值的差异均无统计学意义(P值均>0.05)。Notch4蛋白在不同肺腺癌患者中的IOD值的差异均无统计学意义(P值均>0.05)。结论 Notch受体各亚型中,Notch1、2、3参与了肺腺癌的发生、发展和转移,可能为促癌作用,Notch4的作用不确定。
Objective To investigate the expressions of Notch1-4 proteins in lung adenocarcinoma tissues, and to analyze their correlation with clinical characteristics, and further to discuss its clinical significance in lung cancer. Methods To Collect and stage lung cancer cases of adenocarcinoma and to investigate the parameters of their differentiation degree, tumor size, lymphnode metastasis, clinical staging and so on. The expressions of Notch1-4 proteins in lung adenocarcinoma and adjacent no malignant tissues were detected by immunohistochemistry method respectively, and quantitative analysis of the difference among them were done by Image-Pro Plus 6.0 image analysis software system. Their relationship with clinical pathological factors were further analyzed. Results The IOD values of expression of Notch1、2、3 protein in male patients was significantly higher than that in females patients(P <0.05); The IOD values of expression in patients with the TNM staging Ⅱ and Ⅲ were significantly higher than that in patients with stage TNM staging Ⅰ(P <0.05), The IOD values of expression in patients with stage Ⅲ IOD values were significantly higher than that in patients with TNM stage II (P <0.05); The IOD values of expression in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis (P <0.05); The IOD values of expression in patients with distant metastasis was significantly higher than that in patients without distant metastases (P <0.05);The IOD values differences in patients with different gender, age, degree of differentiation, tumor size were no statistical significance (P> 0.05). The IOD values of expression of Notch4 protein in different lung adenocarcinoma patients was no statistical significance (P> 0.05).Conclusions Notch1-3 protein may play a role in the carcinogenesis, development and metastasis of lung adenocarcinoma, maybe tumor promotion ,while the influence of Notch4 is uncertain.
