摘要:
【摘要】目的 观察血栓素A2受体拮抗剂雷马曲班对嘌呤霉素氨基核苷诱导的足细胞凋亡及nephrin蛋白表达的影响。方法 以条件永生性小鼠足细胞株为研究对象,分为(1)足细胞对照组,(2)足细胞嘌呤霉素氨基核苷(PAN)组,(3)足细胞雷马曲班组,(4)足细胞PAN+雷马曲班组。嘌呤霉素氨基核苷刺激后48h,用流式细胞仪检测足细胞凋亡水平,用实时荧光定量RT-PCR检测nephrin蛋白相对表达量,Western bolt检测nephrin蛋白的表达状况。结果 与对照组及PAN+雷马曲班组比较,PAN组的足细胞凋亡率增加,nephrin表达量下降,具有统计学差异(P<0.05)。结论 TXA2受体拮抗剂雷马曲班可以抑制PAN诱导下的足细胞凋亡,使nephrin表达量上调。
Abstract: Objective To investigate the effect of thromboxane A2 receptor antagonist Ramatroban on podocyte apoptosis which were induced by puromycin aminonucleoside(PAN)and the expression levels of nephrin. Methods The conditionally immortalized murine podocytes were randomly divided into four groups: 1. podocyte control group, 2.podocyte exposed to PAN, 3.podocyte exposed to Ramatroban, 4.podocyte exposed to PAN combined with Ramatroban, The level of podocytes apoptosis was measured by flow cytometry at 48 hours after PAN stimulation. The protein relative expression of nephrin was detected by real time quantitative RT-PCR. The protein expression of nephrin was detected by western blot. Results Compared with normal control group and PAN+ Ramatroban group,the apoptosis of podocytes was increased in PAN group. To the contrary, The protein expression of nephrin was decreased. There was statistical significance(P<0.05). Conclusions Thromboxane A2 receptor antagonist Ramatroban exerted inhibitory effect on the apoptosis of podocytes induced by PAN. The protein expression of nephrin was upregulated.
