摘要:
目的:研究不同剂量氯胺酮对大鼠急性脊髓损伤的影响及其可能机制。方法:SD大鼠60只,随机分为假手术组、损伤组、5mg/kg氯胺酮组、10mg/kg氯胺酮组和20mg/kg氯胺酮组。氯胺酮各组于损伤后4h给予相应剂量的氯胺酮,其余两组以相同的方法给予等量0.9%的生理盐水。采用改良的Allen’s打击法致伤大鼠脊髓。应用BBB(Basso,Beattie and Bresnahan BBB)评分法进行后肢运动功能评价;分别采用黄嘌呤氧化酶法和硫代巴比妥酸法测定血浆超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量变化;同时观察组织形态学变化。结果:10mg/kg和20mg/kg氯胺酮组行为学评分显著高于损伤组;与假手术组相比,损伤组血浆MDA含量显著升高,SOD活力显著降低;与损伤组相比,10mg/kg和20mg/kg氯胺酮组血浆MDA含量显著降低,SOD活力显著升高;损伤组脊髓病理变化较重,可见大量神经元坏死,10mg/kg和20mg/kg氯胺酮组神经元坏死较轻。结论:氯胺酮对大鼠急性脊髓损伤具有保护作用,其机制可能与抗氧化作用有关。
Objective:To study the effects and mechanism of different dose ketamine on acute spinal cord injury in rats. Methords:Sixty Sprayue-Dawleys rats were randomly divided into 5 groups:sham operation group, injury group, 5mg/kg ketamine group, 10mg/kg ketamine group and 20mg/kg ketamine group. In ketamine groups, different dose ketamine was infused in an hour, 0.9% Saline was used in the other groups. Rat models with spinal cord injury were made by a modified Allen’s method. The BBB(Basso,Beattie and Bresnahan) scales were used to assess hind limb neurological function. Superoxide dismutase (SOD) activity, malonaldehyde (MDA) level in the plasm were measured respectively by thiobarbituric acid(TBA) and xanthine oxidase methords. Pathological changes were observed. Results:Behavioral scores of 10mg/kg and 20mg/kg ketamine group was significantly higher than those of injury group. Compared with sham operation group, MDA level in injury group was increased and SOD activity was reduced significantly. Compared with the injury group, MDA level in 10mg/kg and 20mg/kg ketamine group was much lower and SOD activity increased remarkably. In injury group, a large number of neuronal death could be seen, pathological changs in 10mg/kg and 20mg/kg ketamine group were better than those of injury group. Conclusion:Ketamine has neuroprotective effects on acute spinal cord injury, which is possibly related to its antioxidation.
