摘要:
目的 分析外周血赖氨酰氧化酶(LOX)基因rs2278226位点多态性与结直肠癌患者肿瘤转移的关系,并探讨其临床意义。方法 应用DNA直接测序技术检测了2011.1-2015.6于我科住院治疗的164名转移结直肠癌患者(转移组)、218名无转移患者(无转移组)和340名健康体检者(对照组)的外周血LOX基因rs2278226位点的多态性;同时检测了3组人群血清肿瘤标志物癌胚抗原(CEA),CA199,CA50及LOX的水平。 结果 2组结直肠癌患者血清CEA、CA199、CA50及LOX的浓度均显著高于正常对照组,转移组中血清肿瘤标志物CEA、CA199、CA50及LOX的浓度显著高于无转移组(P<0. 05) 。转移组与无转移组CC、CG基因型频率均显著高于对照组(P<0.05);转移组CC基因型频率明显高于无转移组CC基因型频率(P<0.05)。转移组C等位基因的频率为42.4%,无转移组C等位基因的频率为27.5%,明显高于正常对照组(C等位基因频率15.1%),且转移组C等位基因频率显著高于无转移组C等位基因频率(均P<0.05)。Logistic回归分析结果显示CC基因型是结直肠癌患者肿瘤转移的独立危险因素,OR(95%CI)值为1.76(1.3148-2.793) (P<0.05)。结论 结直肠癌患者LOX基因rs2278226位点多态性与肿瘤转移有关。
Objective To polymorphism of lysyl oxidase (LOX) gene in peripheral blood and clinical significance in patients with colorectal cancer. Methods DNA direct sequencing technology was applied to test polymorphism of LOX gene rs2278226 site in peripheral blood in patients with colorectal cancer, and patients were recruited from 2011.1-2015.6 in our department. Then patients were divided into metastasis group(164 cases), non- metastasis group(218 cases), and 340 health examination persons were recruited as control group. Serum CEA, CA199, CA50, LOX were also tested in these 3 groups simultaneously. Results: Concentrations of serun CEA, CA199, CA50, LOX in patients with colorectal cancer were all higher than those in control group, and these indexes in metastasis group were all higher than those in non- metastasis group(P<0.05). Frequency of CC, CG gene types in colorectal cancer patients was higher than that in control group, and and frequency of CC gene type in metastasis group were all higher than those in non-metastasis group(P<0.05). Frequency of allele C was 42.4% in metastasis group and 27.5% in non-metastasis group, which was higher than that in conrol group(15.1%), and frequency of allele C in metastasis group was higher than that in non-metastasis group(P<0.05). Result of Logistic showed that CC gene type was independent risk factor for metastasis of colorectal cancer, and OR(95%CI) was 1.76(1.3148-2.793) (P<0.05). Conclusion Polymorphism of LOX gene rs2278226 site was related to tumor metastasis of colorectal cancer.
