摘要:
目的:探讨穴位电刺激对宫颈扩张大鼠脊髓ERK1/2活化的影响。方法:36只SD大鼠随机分为宫颈扩张(UCD)组(U组)、UCD+经皮穴位电刺激(T组)、与Sham组。U组大鼠手术暴露扩张子宫颈,并在“合谷”与“三阴交”留针,但不给予电刺激;T组大鼠手术暴露扩张子宫颈,取“合谷”与“三阴交”行经皮电刺激;Sham组大鼠手术暴露宫颈不行宫颈扩张,不行穴位穿刺置针。采用免疫组化法观察脊髓ERK1/2磷酸化水平改变(n=4);记录大鼠腹直肌肌电位(EMG)振幅改变(n=8)。结果: 与基础值相比,UCD后U组与T组大鼠EMG振幅增加 (均P<0.05);与U组相比,UCD后各时间点,T组大鼠EMG振幅增加较少(P<0.05)。UCD后U组与T组大鼠脊髓p-ERK1/2标记的阳性细胞数量增加 (P<0.05)。与U组相比,T组大鼠脊髓p-ERK1/2标记的阳性细胞数量较少(P<0.05)。结论: 脊髓ERK1/2信号通路参与宫颈扩张所致的急性内脏痛的信号转导过程,抑制ERK1/2的活化可能是穴位电刺激缓解内脏痛的机制之一。
Objective To explore the effect of Transcutaneous electrical nerve stimulation (TENS) on the activation of spinal extracellular signal-regulated kinases in the acute visceral pain rats . Methods 36 Sprague-Dawley rats were randomly divided into three groups: uterine cervical distension group (U group), Transcutaneous electrical nerve stimulation group(T group), and sham group (S group). U group rats with an abdominal incision and two metal rods inserted through cervical canals, only acupuncture needle into rats with bilateral Hegu and Sanyinjiao, but did not connect Han's Acupoint Nerve Stimulator. T group rats with an abdominal incision and two metal rods inserted through cervical canals, and connect Han's Acupoint Nerve Stimulator on bilateral Hegu and Sanyinjiao. The sham operation group included rats with only an abdominal incision and two metal rods inserted through cervical canals but without distension and TENS. The expression of p-ERK1/2 in spinal dorsal horn neurons was determined by immunohistochemical analysis(n=4) and the electromyographic (EMG) response to uterine cervical distension (UCD) was observed(n=8). Results Compared with the base value, the EMG of the U group was increased significantly between with the T group after UCD(P<0.05). Compared with the U group ,EMG of the T group were increased less at each time point after UCD(P < 0.05). The number of p-ERK1/2 between the U group and the T group after UCD was increased(P< 0.05). Compared with the U group, the number of p-ERK1/2 in the T group was less (P< 0.05). Conclusion The ERK1/2 signaling pathway in spinal dorsal involves in the signal transduction process of UCD-induced acute visceral pain in rats, TENS alleviate pain and the possible mechanism is inhibiting the activation of ERK1/2.
