胃癌患者外周血及肿瘤组织中LMO1、MMP-9、Ecadherin的表达情况及临床意义

摘要:
目的 检测LMO1、MMP-9、Ecadherin蛋白在胃癌患者外周血与肿瘤组织表达情况,对其关系和意义进行探讨。方法 选取经病理确诊的胃癌患者150例为研究组,年龄性别相匹配的健康查体人员50人为对照组,取肿瘤及癌旁组织石蜡标本免疫组化(SP)染色检测肿瘤组织LMO1、MMP-9、Ecadherin蛋白表达情况;用酶联免疫吸附试验(ELISA)方法检测患者手术前、术后1月的外周血中LMO1 、MMP-9、Ecadherin蛋白表达;检测50例健康体检者外周血LMO1 、MMP-9、Ecadherin表达。结果:研究组肿瘤组织中LMO1 、MMP-9的表达明显比癌旁组织增强, Ecadherin的表达明显比癌旁组织降低(P<0.05);LMO1在胃癌组织及外周血中表达存在正相关(均P<0.05);研究组术前外周血中LMO1 、MMP-9水平高于对照组, Ecadherin水平明显低于对照组(P<0.05);手术后研究组患者LMO1、MMP-9水平明显下降,Ecadherin水平明显上升,与对照组差异不明显(P>0.05)。胃癌组织LMO1蛋白表达与淋巴结转移情况有关(P<0.05)。研究组术前LMO1与MMP-9、Ecadherin存在显著正相关关系(P<0.05)。结论:LMO1在外周血表达增高且与胃癌组织中LMO1存在相关,在肿瘤切除后外周血LMO1水平降低。提示外周血LMO1可能作为胃癌肿瘤标志物,监测外周血LOM1对反映胃癌患者的疾病状态有帮助。
Objective: To detect expressions of LOM1, MMP-9, Ecadherin in peripheral blood and gastric cancer tissues, and investigate their relationship and significance. Methods: 150 gastric cancer patients with pathological diagnosis were recruited as research group, and 50 normal cases of health examination with matched age and sex, were recruited as control group. Immunohistochemical assay was performed to detect the expressions of LOM1, MMP-9, Ecadherin in gastric cancer tissues and normal mucosa, while their expressions in peripheral blood was tested with enzyme linked immunosorbent assay(ELISA)both before and one month after operation. Also their expressions in peripheral blood of the control group were tested with ELISA. Results: Expressions of LMO1, MMP-9 in in gastric cancer tissues were remarkably higher than those in normal mucosa, while expression of Ecadherin was obviously lower in gastric cancer tissues than that in normal mucosa(P<0.05). Significant positive correlation was found between LMO1expression in tumor tissues and its expression in peripheral blood(P<0.05). Before operation, levels of LMO1 and MMP-9 in peripheral blood of research group were higher than those in control group, while the level of Ecadherin was relatively lower (P<0.05). After operation, expressions of LMO1, MMP-9 in peripheral blood of research group decreased significantly, whereas the level of Ecadherin increased greatly (P<0.05), and these indexes had no significant difference with those of control group(P>0.05). Expression of LOM1 protein in gastric cancer was related to lymphatic metastasis(P<0.05). Significant correlations were found between LMO1 and MMP-9, LMO1 and Ecadherin in peripheral blood in research group before operation (P<0.05). Conclusion: Significant correlation was found between LMO1 in cancer tissues and the up-regulation of LMO1 in peripheral blood. That the level of LMO1 in peripheral blood decreased after operation suggests LMO1 in peripheral blood can be used as tumor markers for gastric cancer. Detection of it can reflect the disease state of patients with gastric cancer.
