摘要:
目的 检测Livin在子宫内膜癌组织中的表达与临床病理分期的关系,研究干扰Livin表达对子宫内膜癌细胞侵袭转移能力的影响。方法 应用荧光定量PCR法检测Livin mRNA在子宫内膜癌组、非典型增生组和正常内膜组中的表达,分析其差异性表达与子宫内膜癌临床病理分期的关系。将Livin的小分子干扰RNA(Livin-siRNA)和阴性对照序列(Livin-NC),应用脂质体法转染Ishikawa和HEC-1-A细胞,转染后48h,Western Blot法检测各组细胞Livin蛋白的表达,Transwell小室侵袭和迁移实验观察细胞穿膜能力的变化。结果 Livin mRNA在子宫内膜癌组表达上调(P<0.01),Livin在子宫内膜癌中的表达与肌层浸润深度和病理分级有相关性(P<0.05),与患者年龄、FIGO分期和淋巴结转移无相关性(P>0.05)。转染后Livin蛋白表达下调(P<0.05),Transwell小室侵袭和迁移实验显示,干扰Livin表达后穿膜细胞数明显减少(P<0.01)。结论 Livin在子宫内膜癌组织中表达上调并且与肌层浸润深度和病理分级有关,下调Livin表达可抑制子宫内膜癌细胞的侵袭和迁移能力。
Objective To examine the relationship between the expression of Livin in endometrial carcinoma and clinicopathological staging, and study the effect of Livin on the invasion and migration of endometrial carcinoma cells. Methods The expression of Livin mRNA in endometrial carcinoma, atypical hyperplasia and normal endometrium was detected by fluorescent quantitative PCR. The relationship between the expression of Livin mRNA and clinicopathological staging of endometrial carcinoma was analyzed. Livin-siRNA and negative control sequence (Livin-NC) were transfected into Ishikawa and HEC-1-A cells by liposome method. Western Blot was used to detect the expression of Livin protein in each group after 48 hours. Transwell chamber invasion and migration experiments were performed to observe the changes of cell membrane penetration ability. Results The expression of Livin mRNA was up-regulated in the endometrial carcinoma group (P<0.01). The expression of Livin in endometrial carcinoma was related to the depth of myometrial invasion and pathological grade (P<0.05), not related to patient age, FIGO stage, and lymph node metastasis (P>0.05). Livin expression was down-regulated after transfection (P<0.05). Transwell chamber invasion and migration experiments showed that the number of transmembrane cells was significantly reduced after interference with Livin expression. (P<0.01). Conclusion The expression of Livin in endometrial carcinoma is up-regulated and related to the depth of myometrial invasion and pathological grade. Down-regulation of Livin expression can inhibit the invasion and migration of endometrial carcinoma cells.
