摘要:
目的 研究自噬相关基因及蛋白在慢性阻塞性肺疾病患者肺组织标本中的表达。方法 标本取自45例因肺部占位病变行肺叶切除术患者的肺组织,按术前肺功能分为非COPD组、COPD轻度组、COPD中度组、COPD重度组。所取肺组织用免疫组化染色检测自噬蛋白LC3B的表达,用Western Blot和real-time PCR检测肺组织标本中自噬相关基因Atg5、Beclin1 及自噬相关蛋白Atg5、Beclin1、LC3B的表达。结果 Real-time PCR分析结果显示Beclin1在COPD轻度组、中度组及重度组mRNA表达均显著高于对照组(其中轻度组与中度组P<0.01,重度组P<0.05),Atg5在COPD轻度组和中度组mRNA显著高于对照组(其中轻度组P<0.05,中度组P<0.01),重度组没有统计学意义(P>0.05)。Western blot分析结果显示Beclin1在COPD轻度组、中度组及重度组蛋白表达均显著高于对照组(其中轻度组和中度组P<0.01,重度组P<0.05),Atg5在COPD轻度组和中度组蛋白表达显著高于对照组(其中轻度组P<0.05,中度组P<0.01),重度组没有统计学意义(P>0.05)。LC3B在COPD轻度组、中度组及重度组蛋白表达均显著高于对照组(其中轻度组和中度组P<0.01,重度组P<0.05)。 结论 自噬相关基因及蛋白在慢性阻塞性肺疾病的进展过程中表达有所增加,提示可能与疾病的发展有关。
【Abstract】 Objective To investigate the expression of autophage-related gene and proteins in the lung tissue of patients with chronic obstructive pulmonary disease (COPD). Methods Pulmonary tissues were obtained from 45 subjects who were undergoing single or bilateral lobectomy or wedge resection for lung occupying lesions.They were divided into non-COPD group, mild COPD group, moderate COPD group and severe COPD group according to preoperative pulmonary function. The expressions of autophagy genes Atg5, Beclin1 and autophagy proteins Atg5, Beclin1 and LC3B in lung tissue were measured by Western Blot and real-time PCR. The expression of autophagy protein LC3B was measured by immunohistochemical staining. Results Real-time PCR analysis showed that the expression of Beclin in COPD mild group, moderate group and severe group were significantly higher than those in the control group (mild group and moderate group vs control group P <0.01, severe group vs control group P <0.05). The mRNA levels of Atg5 in mild group and moderate group were significantly higher than those of the control group (mild group vs control group P <0.05, moderate group vs control group P <0.01), but there was no significant difference between severe COPD and control group (P <0.05). Western blot analysis showed that the expression of Beclin in COPD mild group, moderate group and severe group were significantly higher than those in the control group (mild group and moderate group P <0.01, severe group P <0.05), The protein levels of Atg5 in mild group and moderate group were significantly higher than those of the control group but there was no significant difference between the severe COPD and control group (P >0.05). The expression of LC3B in COPD mild group, moderate group and severe group was significantly higher than that in the control group (mild group and moderate group vs control group P <0.01, severe group vs control group P <0.05 ). Conclusion Autophagy genes and proteins are increased in the progression of chronic obstructive pulmonary disease, suggesting that autophagy may play an important role in the progress of COPD.
