摘要:
【摘要】 目的:探讨运动试验中血压变化与高血压患者靶器官的损伤和血浆及外周血单个核细胞(PBMC)中microRNAs表达变化的关系。方法:筛选90例高血压患者,根据运动试验中患者血压升高水平,分为运动性高血压组与普通高血压组。选取普通健康人20例作为正常对照组,记录3组患者的临床资料,检测患者靶器官损伤相关指标,并使用实时荧光定量PCR检测三组患者血浆及PBMC中microRNAs的表达水平。结果:运动性高血压组的与普通高血压组及正常对照组比较靶器官损伤指标差异均有统计学意义(P<0.05);运动性高血压组血浆及PBMC中miR-145、mi-R-21表达与普通高血压组比较,差异均有统计学意义(P<0.05),且普通高血压组与正常对照组比较,差异均有统计学意义(P<0.05);三组血浆中miR-143表达差异无统计学意义(P>0.05),但三组PBMC中miR-143表达差异具有统计学意义(P<0.05)。结论:提示运动性高血压可以被视为评估靶器官损伤相关指标血压与靶器官损伤之间联系的一个重要因素,同时发现microRNAs在运动性高血压病人与普通高血压病人及健康人的血浆和PBMCs中的表达不同,在某种程度上反映出高血压疾病的状态,有望为高血压的预防及诊疗提供新的分子标志物和研究方向。
【Abstract】 Objective To explore the correlation of blood pressure(BP) response in exercise testing in patients with subclinical target organ damage and expression level and clinical significance of microRNAs in hypertension patients in plasma and peripheral blood mononuclear cells(PBMC). Methods A total of 90 hypertension patients were divided, according to levels of blood pressure in exercise testing, into common hypertension group(n=55) and exercise hypertension group(n=35), selected 20 Normal healthy people as normal control group. Clinical data and target organ damage related indicators were recorded. Expression of microRNAs in plasma and PBMC in different groups was detected by real-time quantitative PCR . Results exercise hypertension group were different from common hypertension group and normal control group in target organ damage related indicators(P<0.05); exercise hypertension group were different from common hypertension group and normal control group in expression of miR-145、miR-21 in plasma and PBMC, and common hypertension group were different from normal control group. There was no significant difference in expression of miR-143 in plasma between 3 group (P>0.05), but significant difference was noted in expression of miR-143 in PBMC between exercise hypertension group and common hypertension group (P<0.05), and between common hypertension group and normal control group (P<0.05). Conclusion exercise hypertension can be regarded as an important factor in assessing the relationship between hypertension and target. At the same time, it is discovered that microRNAs have different expression in plasma and PBMCs of patients with normal hypertension and exercise hypertension, which reflects the state of hypertension disease in some degree and provides new molecular markers and research direction for the prevention and diagnosis of hypertension. 【Key word】hypertension; Exercise test; MicroRNA; Vascular smooth muscle cell(VSMC)
